I haven't done it, but the 250 is about 1 1/2" longer and will require moving the radiator forward. The bellhousings are different, but the tranny pattern is the same, so reusing the tranny is possible. Clutch linkage may need to be reworked some, and the motor mounts are totally different. This swap is about the same in dificulty/expense as doing a V8 swap.

Gearhead is right, It's a major job to gain 20 cid which is almost nothing.
I would recommend finding a 261" for your pick up. They can be built to 278" & will bolt right in etc.
Good luck, John M., #3370

i have just finished doing this . my 39 suburban is on a frame from the 50's . not sure of the year the motor is a later Maybe '70 250 .the hard part was switching the crossmember to accomodate the bell housing difference.a lot of rivets to chisel grind and beat off .i was fortunate to have this come out of a 55 truck that had front side mount adapters made to attach to the motor side mounts .a sort of sideways u-shaped piece of metal bolted to the frame bolted top and bottom .home made but they work. the clutch worked the same . in my truck i had to mount the radiator way up in the front directly behind the grill but this was necessary regardless because of the older body length.
